Can this be removed without anything else being done or taken off? If so how hard is it to put back on?

Which coolant line? The black one that goes from the heater core to the water pump?

The off white colored one right inbetween the intake and the oil cap

The overflow tube can be removed by itself, nothing else has to come off. Make sure the coolant is cool when doing this.

Edit: You may have to clear the air out of the system after doing it.

Let the car get to operating temp, with the t-stat open. Where the water pump is, there is a black tube with a brass fitting on top. Take that off and air will come out of the system, Once a steady stream of coolant comes out, all the air is purged.
